frustrating...
Yesterday I was doing a very long experiment, basically I worked the whole day without any rest. I need to phenotype my conditional KO mice, and with 3 mice per group, I have 9 mice to analyze.
In the normal situation, analyzing 9 mice is not too big a deal. However, this is my first time screening, so to be more careful, we collected 5 different tissues including bone marrow, spleen, payer's patch, peritoneal cells and thymus. Therefore, yes, I have 45 cells to count, which took me almost 3 hours!
I know I am not phisically strong enough to work so long right now, but this is a very important experiment and there's no way I can stop and come back to work the next day. When I finished counting cells and started to stain with antibodies, it was almost 6 pm. I was exhausted. And here comes the tragedy. After I washed the cells and centrifuge them, cells were frozen!!!!!
I checked the centrifuge, the temperature setting was at -2 C. It is very wired because I was the only one using the machine yesterday, and I didn't adjust the setting at all. It is possible that somebody accidently touched it and wasn't aware of, and of course, I might be the "somebody".
Anyways I was shocked for couple seconds and then immediately put the cells into 37C waterbath trying to rescue them. But I know once cells are frozen, they might all be lyzed. I sadly kept staining and fixed the cells. When I left the lab, it was almost 9pm.
It is a very bad idea to do a very long experiment alone. Although my boss helped me to collect the tissues in the morning, it is still very tiring. I was sad and frustrating this morning when I told my boss what happened this morning in the lab meeting. Other people in the lab told me the centrifuge is old and the temperature setting sometimes just goes crazy. Oh well, I guess I am just unlucky, the machine knows I am new to the lab.
Since cells are still there, I analyzed them by flow cytometry this afternoon. Surprisingly, they looked OK!
Maybe I am not that unlucky afterall. Haha.
Conclusion: Never ever do any experiment alone during after hour!!!!!!

雜記
1. 網誌右下方放了可愛的貓咪溫度計，按一下兩個圈圈那個地方會顯示houston現在的溫度，貓咪也會跟著天氣呈現不同的姿勢，很可愛喔。不過最近幾天houston都是heat advisory，一整天超過100F以上的時間有好幾個小時，真的快熱爆了！
2. UT daycare的價錢是一個月$700，而且有長長的waiting list，不知道我們到底排不排的到呢？
3. 提不起勁唸書，很苦惱，而且上兩星期做的實驗都是genome typing，覺得自己很廢，還好明天要做一堆實驗，希望有好data。
4. 好像開始有點水腫了，晚上比較明顯。
5. 想要再養一隻貓跟kobe作伴。
6. 最近購物慾很旺盛，但衣服鞋子什麼的，喜歡也不能買（不能穿或穿不下），只好把目標轉移到跟身材無關的東西上，很辛酸啊！
7. 雖然不需要你們欺哄我說我懷孕沒有胖太多，但基本上孕婦還是不喜歡討論體重話題的，謝謝大家。

Lucky
I guess I am lucky in a way.
One of our friends is also pregnant almost at the same time as I. But she has been in hospital since 3 weeks ago because of bleeding and constraction. Her doctor recommends her to stay in the hospital until 37 weeks, which means 7 weeks more. I can't imagine how difficult it will be if I have to go through the same torture.
Thank God I have a healthy pregnancy and a healthy baby. Although my baby doesn't have love from father yet, at least we both are doing well.
Right now my only worry is that Udon may not get along with the baby, or I should say, Udon may want to "play with" (or just "play") the baby all the time. Hopefully Udon will be mature enough to become a big brother to her. BTW, Udon is right now 62 pounds!!! Soooo big!!!
I still don't have any idea about her name, any suggestion?

寄信真方便
之前幫朋友訂衣服，結果訂錯尺寸，其實早就要寄回去換了，可是每天出門回家的時間都跟郵局上班很不合，於是一拖拖了兩個禮拜，昨天忽然想到，要是再不退，搞不好會換不到尺寸，所以打定主意想說要早起去一趟郵局。但好像是因為我沒有大信封可以裝吧，居然還是莫名其妙的直接把衣服帶去學校了。
快中午的時候經過學校的郵件室，順手拿了一個flat rate的priority信封，想說應該放的進去兩件衣服，拿是拿了，但本來我是小氣的想寄平信，畢竟郵資要自己負擔。上網查了一下郵資，發現其實可以刷卡印郵票出來，而且只要輸入收件人寄件人的資料，就可以印出整張包含郵資還有條碼的詳細單據，接下來貼到信封上就可以投到郵筒了呢（要是再懶一點，還可以要求郵差去你家或公司取件喔）。可惜這個服務不適用於平信，於是本人的懶惰戰勝三元美金，最後還是用priority mail寄出了。
老實說，郵局離我們家其實很近，但想到只要有印表機，隨時都可以印郵資就覺得非常方便，唯一的麻煩就是要有秤囉，還好實驗室都有電子秤，不擔心不擔心。只是這種懶人的便利措施，我怎麼會到現在才想到要用啊？真是愧對於自己。不過話說回來，要寄國際包裹可能也不行這麼懶，因為還是得要填海關單子那些的。

p.s. 學校的郵件室可以幫你秤還有計算郵資，但只有自動販賣郵票機，不能直接跟他買郵資，所以還是有些不方便。well，也是可以買很多張41 cent 的郵票貼滿整個信封啦，只是有點累人。
